Dr Liang is currently a project director at the Lady Davis Institute of the Jewish General Hospital, as well as an Associate Professor in the Department of Medicine, McGill University. He is well recognized for his earlier work on HIV-1 genomic RNA packaging and particle assembly, with a focus on elucidating the mechanisms behind the selective packaging of the dimeric viral genomic RNA and deciphering the molecular details of viral Gag protein multimerization. His group has since expanded its scope of research toward understanding the complex interactions between HIV-1 and the host cells by taking the proteomic and genomic approaches to identify cellular factors that potently modulate HIV-1 replication and to elucidate their mechanisms of action. Dr Liang’s group is also actively involved in HIV-1 vaccine research and HIV-1 latency study.
Major Research Activities
Le groupe du Dr Liang s’intéresse à la compréhension des interactions entre le VIH-1 et les cellules hôtes. Un aspect de ses recherches implique la découverte des facteurs de restriction de l’hôte, qui inhibent l’infection par le VIH-1, et la compréhension des mécanismes moléculaires sous-jacents. Les facteurs cellulaires à l’étude présentement comprennent l’antigène 2 des cellules stromales de la moelle osseuse (BST-2, aussi appelé tétherine), qui inhibe la libération de virus de la surface cellulaire, l’APOBEC3G et MOV10, qui ciblent toutes les deux les complexes protéiques de l’ARN du VIH-1 et compromettent la transcription inverse virale, les protéines transmembranaires induites par l’interféron (IFITM), qui inhibent l’entrée du virus, et la protéine MxB, qui empêche l’importation nucléaire du VIH-1. Son groupe s’intéresse aussi au rôle des hélicases cellulaires à divers stades de réplication de l’ARN du VIH-1. Des études connexes ont conduit à l’identification de plusieurs hélicases qui régulent la transcription, la traduction, l’encapsidation et la transcription inverse de l’ARN viral. Les principales découvertes du laboratoire du Dr Liang comprennent l’activité anti-VIH-1 des protéines IFITM et MxB. Son groupe applique aussi l’outil de modification génomique du CRISPR/Car9 pour étudier la latence du VIH-1. Le but de ses recherches consiste à faire progresser notre compréhension de la pathogenèse du VIH-1 et découvrir un remède contre le VIH.
